چکیده

ZR-75-1 human mammary cancer cells can be grown in serumfree medium in an estradiciand tamoxifen-sensitive manner. Growth occurs both in monolayer and suspension culture, al though serum is necessary during the initial plating of the cells in monolayer culture. Optimal stimulation of proliferation by estradiol occurs at 10~8 and 10~9 M. Tamoxifen alone weakly stimulated growth in a dose-dependent manner up to 10~7 M, but inhibition was observed at 10~6 M. This inhibition was not seen if the cells were cultured in 10% dextran:charcoal-treated fetal calf serum. This difference is ascribed to tamoxifen binding components in the serum. In the absence of serum, tamoxifen (10~6 M) abolished the proliferative effect of estradiol at all concentrations of estradici below 10~7 M. Higher concentrations of estradiol partially over came the tamoxifen effect. Consistent antiestrogenic effects of tamoxifen required an estradiol:tamoxifen ratio of 1:1000.

P-195: Thymoquinone Increases Efficacy of Tamoxifen Induced Apoptosis in Human Breast Cancer MCF-7 Cells: In Vitro

Background: The objective of this study is to evaluate combined effect of Thymoquinone (The main active component of black seeds) with Tamoxifen drug on apoptosis of human breast cancer MCF-7 cells (Noninvasive human breast cancer cell line, estrogen receptor positive). Materials and Methods: The human breast cancer MCF- 7 cells were treated with Tamoxifen (2 μM) alone or in combination with Th...

Effect of 17-? Estradiol on the Expression of Inducible Nitric oxide Synthase in Parent and Tamoxifen Resistant T47D Breast Cancer Cells

Indirect evidence suggests that estrogen is involved in the etiology of breast cancer. Estrogen is also thought to modulate nitric oxide (NO) in human breast tumor tissue via regulation of inducible nitric oxide synthase (iNOS). Objectives of this study were to determine whether estradiol (E2) affects iNOS expression level in breast cancer cells and to study the effect of various concentrations...

Antiproliferative effects of flavonoid fractions from Calendula officinalis flowers in parent and tamoxifen resistant T47D human breast cancer cells

The risk of human breast cancer is concerned to cumulative exposure of the breast cells to endogenous estrogens. Strategies aiming at reducing the production of estrogens may be useful for the prevention of estrogens-related breast cancer. Several natural products with plant origin have the potential value as chemo-preventive or therapeutic agents in cancer. Flavonoids, the natural polyphenol c...
